I believe you can use multiple readyboost drives but I haven't tried that. I've been using a 2GIG USB drive for readyboost on my laptop for over a year. It does help noticably.

Vista checks your USB drive to see if it is fast enough to use for readyboost. A lot of older USB drives fail and can not be used. Lots of newer drives indicate readyboost ready on the package. On older drives the only way to tell is to try them and see.
