- How large are the .DBF/.FPT/.CDX?
- Are they stored on the W2K8 server, or on the Vista workstation? If the former, can you test with copies of the data on the workstation/local machine?
- Is there any real-time antivirus scanning occurring?

How about some basic information about the computers:
- Processor type and speed, and installed RAM?
- How much RAM is actually being used? Is there plenty of free RAM? How hard is the CPU(s) working/percent utilization?

Anything else you can think of that might be considered "unusual" in your environment?

With Vista clients on a W2K8 network (or Vista peer-to-peer), by default the networking may be SMBv2. It might be worth doing a test with networking forced to SMBv1: message#1332135
