I've worked with both DotNetNuke and Mere Mortals .NET. In my opinion, DotNetNuke is nice if you want already built functionality. There are a lot of 3rd party applications for DotNetNuke, called modules in DotNetNuke, that you can get for free or for a very low cost. For example, there are forums modules, calendar modules, photo gallery modules, video modules, etc. that are very good and very effective for the price.

If you plan on building most of your functionality from scratch, I think Mere Mortals is the best way to go.
