I've found that a lot of anti-virus software can cause this problem depending upon how you having things setup.
If you have your sourcesafe database installed on another machine (like a server or something) - and you have your anti-virus software setup to scan networked drives, then you will have this problem. What I've done to get around it is simply change my anti-virus software configuration to NOT scan networked drives and it fixes the issue. Naturally you'll want to be sure the networked machine has it's own anti-virus software on it.
