I think the App Store is only part of it. The iPhone was successful for the first year before the App Store was launched. The experience using an iPhone was significantly different than any previous smartphone, at least to a lot of people. What the iPhone did is be the first smartphone that really successfully implemented the right software for use with a handheld touch screen, and not requiring a stylus. Having an iPod/iTunes functionality built-in to the first iPhone also helped in early adoption.
