I have tested TSPlus on Windows Server 2003, 2008, and 2012. It will be compatible with all versions I am sure. 64 bit is best because of access to lots of RAM.

TSPlus can run your app either as "virtual" desktop app (not difference to what the user perceives i.e. has a desktop icon and runs the app which appears on the taskbar) or as an HTML5 app within a modern web browser.

Number of users a server can handle will be dependent, obviously, on what the user+app is doing and what the server specs are. Each user will be creating their own session on the server (happens very quickly) and thus will use whatever RAM and CPU cycles the app would consume if run locally - but now it happens on the server along with all other users. So you have to estimate what an average user session will consume in terms of RAM and CPU cycles bearing in mind most apps do not run max CPU all the time anyway but will probably spend a good deal of time consuming very few CPU cycles.

So what server spec do you need? Well more RAM is better than less. More CPU's are better than fewer. And a SSD is much better than a HDD. Get a server you can upscale as required.
