You have to at least stop the Web server, since it is caching your DLL. Go to Control Panel and open the Services program item. On your services list, there shoud be a service named World Wide Web Publishing Service. Stop it and you will be able to recompile your DLL unless you are using Transaction Server. O f course, you'll have to restart the Service to use ASP.
