Yes, and its more than just copying files that is needed - there are updated shared components and regoistry entries as a part of SP3. the easiest thing to do is to create a fresh runtime install as part of your update disk and have it run; if the runtime is up to date, nothing will be changed, and if the files need updating, they'll get updated.
