You can block access to the floppy in several ways, but in order to prvent boot sector virus unfections, you'll need to set up the system to disallow boot from a floppy - a BIOS function. Some more recent BIOSes will also allow you to boot from a CD, and the same applies - to protect against boot sector viruses, don't allow your system to boot from CD, either.
TWEAKUI can prevent the user from accessing selected drive letters; it can be overridden if you know about TweakUI, but if it happens, the user had to do something to let it happen.
I'm very happy with McAfee's VirusScan/Total Virus Defense product; in conjunction with blocking boot from floppy, it's been very successful at keeping my sites clean, as long as the site kept the AV software up to date. NAV works well, too. And there are others. I'd check PCMagazine and PCWorld; both have done reviews of AV products within the past 3 months.
