Yes, in addition to the logical separation of the business layer from the presentation layer, how you you go about physically isolating the layers? For example, you decide to deploy your business layer on a different system from both the database and the UI systems. What communication mechanism should (would) you use? DCOM is an obvious choice, along with XML (plus SOAP). What are the limitations of each (latency, complexity, security, etc)?

Have you deployed any apps. using any of these technologies and if so, what worked well and what would you do differently the next time around? How is maintenance handled?
