It is possible to get an exclusive lock on a table. You can use the TABLOCKX hint within a transaction. The lock will be held for the length of the transaction.

BEGIN TRANSACTION
SELECT * FROM table WITH (TABLOCKX)
...
COMMIT TRANSACTION
