You can from a DOS prompt or in your AUTOEXEC.BAT
SUBST X: C:\folder
Note that if you're running W95/W98 that folder name must be a DOS 8.3 type name, not a long name. This is true for putting it in your AUTOEXEC.BAT, anyways.
If you want an icon to show up put this file in the folder:
AUTORUN.INF
[AutoRun]
ICON=c:\icons\youricon.ico
