Presumably, this is in an application. So you control the menu. If you don't want the menu to use a particular shortcut for an item, remove that shortcut in the Options dialog for the menu item. Having done that, the shortcut keystroke is available to you.

Having said that, I recommend that you put the items for which you want shortcuts on the menu and take advantage of the functionality that's already there. Why recreate the wheel? Add a menu item for Save, give it CTRL+S as a shortcut and then add the appropriate call for what happens when the user chooses the Save item.
