You haven't really given enough detail to answer your question...
but in general, if you are developing for multiple resolutions/colour depths, then always develop to the lowest level.
Ie,
Set your system up to 800x600 256 colours.. then set up all the colours.
If the user wants to run at a higher colour depth than this, the system won't have any problems finding an equivalent in whichever pallette is currently available.
