You really haven't made a mistake, since you can easily convert FAT to NTFS, but not the reverse. As such, common wisdom seems to be that if you are not sure, pick FAT first...

You can convert FAT to NTFS using the CONVERT command line. (There is no GUI for this.) Here's an example:

CONVERT f: /FS:NTFS

If you are converting from a partition with no files open, you may not even have to reboot afterward. If it isn't possible, you'll be prompted to reboot first, and the conversion is done during the boot process before other services are running that keep files open.

Of course, you should backup any drive before attempting this, just in case.
