There are both WinAPI functions and methods of the WMI controls which will enumerate all systems holding an active handle for shared network resources, including shared files, for a specific machine; there are a number of downloads and FAQ entries here on UT which use the Windows Network API services in the Platform SDK, and plenty of WMI examples in the MSDN, nearly all of which are implemented in C++, C# or VB. It'd be reasonably simple to use these methods to detect the number of users that use some common file to find out how many users are running.
