Have you looked at server load, or other server performance montioring? Perhaps your server can't handle the load, or you have a method that is taking a lot of processing power, or something that is hitting the database hard.
True this, see if you can reproduce the error, w/ just an HTML page, testing.html or something.. Reload it several times, perhaps right a little application to hit it every 20 seconds, and download the HTML. That will be the first thing, that will let you begin seeing if the problem is with server load or with ASP.NET etc.
