Get 2-3 SQL Server books - read them from cover to cover. Get both SQL Admin and Development type of books.
Understand how to administer the database and server
Understand the users and roles and how you want to incorporate them in your app.
Understand how to search for problems in the server - sql profiler, management/activity, etc.
Understand how to backup - restore.
Determine how you will run a testing and production db - how will you connect?
Learn about maintenance on the db?
What about a report database (warehouse)?
Understand views, stored procedures, user defined functions.
Understand indexing on the tables - clustered/non-clustered - benefits (why you would use each)
Design your database
Create your database
Use names for objects in the database that make it easy to visually administer - tblap... (Accounts Payable tables), tblpo... (Purchase Order table)

After you've got all this - then start designing your app.
