I found the following quite reliable on Win2K:

http://www.morphasys.com/autodefrag/Files/v1.2/AutoDeFrag.exe

Example AT command:

at 21:00 /interactive /every:M,T,W,Th,F,S,Su "autodefrag"

With no arguments passed it will defrag all drives, the optional argument allows you to specify a single drive to be defragged.
