This is more or less what I have done, especially when I worked at Bata Shoe Organization.

Note that it is safer to do an XCOPY or similar when nobody is using the data. In other words, the backup that I did at midnight, I considered my main backup; I did another backup at noon, just in case, but I didn't keep it in the long run.

Download #10099 might help you implement the backup for different cases; just put all your options (folder to be backed up, files to be excluded, compression command, etc.) into a text file.
