Up until windows98, MS used to ship an excellent program to remove spyware and it was extremely easy to use: format.com

OK, seriously, you should get 2 programs because one alone is very likely to miss a number of parasites (may the fleas of a thousand cammels nest between the legs of anyone involved in creating spyware). With 2 programs, the chances of missed infections decrease dramatically.

After you're supposedly clean, install firefox and use that instead of IE. It's less succeptible to infection.
