Sorry, away on fieldwork.
1. When starting IS12 (I have IS12 Express I should say), optionally use a rightclick to 'Run as Administrator' if you intend to test the package from the application (well just in case, anyway).
2. What is necessary is under 'Build your release' to set the 'Required execution level' to Administrator. Under UAC in Vista, even if you are an administrator, Installshield will run with standard user privileges until it needs administrative privileges. In respect of an installer, this does not allow registry settings to be made. Setting the required execution level in advance means that Installshield will continuously have administrative privileges on the user's machine without them having to request this.
Having said this, your problem is probably quite another one...
