You could call strtran twice:
REPLACE mymemo WITH STRTRAN(mymemo,'my text','new text')
REPLACE mymemo WITH STRTRAN(mymemo,UPPER('my text'),'new text')
but for case of mixed case you should write custom function/procedure/method.
