Can you check the current activity to see if any one else is in it?

I'm not quite sure how to do it but cant you set the database offline.

something like;
ALTER DATABASE databasename SET OFFLINE WITH ROLLBACK IMMEDIATE

check it out in Books online I think it will throw anybody and everybody out of the database.

When I Try to take the backup of the database either from the Enterprize Manager or the Query Analyzer, it fails giving the error that "the backup operation is falied... see the errorlog for details". when I see the errorlog, it was recorded that the backup was falied due to "File Access Denied".
